Peripartum fibroblasts, when initially cultured in monolayers, exhibit a rapid rate of proliferation. Fibroblasts from different tissue locations differ in their proliferative responses to serum and growth factors. Fibroblasts from the liver, lung, and dermal (skin) regions of the animal proliferate at a lower rate than those from most other locations. When cells from different tissue locations are co-cultured, all cells proliferate at the same rate. Fibroblasts from the cervix, endometrium, fallopian tube, and amniotic fluid proliferate at higher rates than those from the other locations. Fibroblasts from the uterus proliferate at higher rates during pregnancy and during the peripartum period. Peripartum fibroblasts divide at higher rates in the presence of serum but do not undergo more than one or two complete cell cycles. Growth factors also affect the rate of proliferation of peripartum fibroblasts. Fibroblasts from the fallopian tube, endometrium, and amniotic fluid are stimulated to divide by transforming growth factor-beta. In contrast, transforming growth factor-beta has little or no effect on the growth of fibroblasts from the skin and the liver. Peripartum fibroblasts from the fallopian tube, endometrium, and amniotic fluid can be stimulated to divide by epidermal growth factor, platelet-derived growth factor, or both. In addition, these fibroblasts are also stimulated to divide by low concentrations of serum. These observations suggest that the trophic factors in serum may play a role in regulating growth of peripartum fibroblasts and may provide clues to the control of the rapid growth of these cells during the peripartum period.
